Copper-catalyzed preparation of triazoloindoles and their utilities as alpha-imino rhodium carbene

 

Abstract

We have developed an efficient method for the preparation of triazoloindoles (3) from ortho-alkynylanilines (1) and azides (2) via copper catalyzed alkyne azide cycloaddtions (CuAACs) by stabilizing 5-copper-1,2,3-triazole intermediate through the intermolecular and intramolecular coordination. Tosyl azide not only worked as a component, but also a reductant in this CuAAC reaction. Triazoloindoles (3) were further disclosed as precursors of a-imino rhodium carbene and were applied in the synthesis of a series of indole alkaloids.

Profile. Yanguang Wang was born in 1964 in Shanxi Provence, China. He received his B.S. (1985), M.S. (1988) and Ph.D. (1993) degrees from Lanzhou University. He did the post-doctoral research with Professor Henry N. C. Wong at the Chinese University of Hong Kong during 1997~1998. Since 1994, he joined Zhejiang University, where he was promoted to be a full professor in 1998. Now he is the head of Chemistry Department of Zhejiang University, the president of the Chemical Society of Zhejiang Province, and the member of the editorial board of Chemical Journal of Chinese University and Chemical Research in Chinese University. His major research interests include new synthetic methodologies of heterocyclic and carbocyclic molecules as well as total synthesis of natural products and their analogues.
