Prerequisites Course Contents Fundamentals of Biochemistry: This section will deal with the experiments which can be 20used to determine the bio-molecules both quantitatively and qualitatively. Experiment related to Spectro-photometry Beer Lambert Law. Experiment describing the preparation of standard plot for model protein such as bovine serum albumin (BSA). Quantifying the unknown protein using spectro-photometric measurements by UV adsorption, Lowry method, dye binding method, Bicinchoninic acid (BCA) method. Analysis of the presence of glucose and quantifying the concentration using Dinitrosalicylic acid (DNSA) method. Enzyme activity, Enzyme kinetics, Starch conversion to glucose by salivary amylase. Paranitrophenyl 13Dglucopyranoside (pNPG) assay for 13glucosidase. Analysis of proteins by SDSPAGE. Biochemical Engineering Bio-separations and Bioprocesses: Experiments related to 20upstream processing and downstream processing. Fermentation and microbial mass culture. Downstream processing; harvesting of cell mass and extraction of protein/enzyme (adsorption, filtration}, cell sonication, precipitation, column chromatography, column re-generation, affinity column preparation, purification, protein and enzyme activity measurements, SDSPAGE analysis, data compiling. Ethanol Fermentation. Bioreactor operation (25 Its), tangential/cross flow filtration for cell harvesting, computer simulation, mass transfer phenomenon and cell growth kinetics. Topics
